This project aims to develop a novel balloon catheter decorated with a continuous layer of nanoneedles for advanced lipidomics analysis during angioplasty. The catheter functions as a standard balloon angioplasty device while also enabling the nanoneedles to noninvasively sample lipid profiles from atherosclerotic plaques and the surrounding vessel during the procedure. The sampled lipids can subsequently be analysed in bulk or through spatial lipidomics to generate a detailed and personalised lipidomic profile for guiding patient-specific treatment strategies. The project will focus on: 1) manufacturing the nanoneedle-decorated balloon catheter to medical-grade standards; 2) testing its deployment in an ex vivo porcine model of balloon angioplasty; 3) evaluating the catheter’s lipidomic profiling capabilities in both the porcine model and clinical samples of atherosclerotic plaques. This innovative approach integrates therapeutic and diagnostic functions, paving the way for more effective and personalised treatments for atherosclerosis.
